Once-Weekly Semaglutide 2.4 mg in Adults with Overweight or Obesity

Wilding JPH, Batterham RL, Calanna S, et al. · New England Journal of Medicine

The trial that reset expectations for pharmacologic weight loss: 1,961 participants, 68 weeks, and a 14.9% mean reduction in body weight versus 2.4% on placebo.

Key findings

  • 86% of treated participants lost ≥5% of body weight
  • One third lost ≥20% of body weight
  • Improvements across waist circumference, HbA1c, CRP, and lipids
  • GI adverse events led to 7% discontinuation

Limitations

Intensive lifestyle counseling was provided to both arms; results may overstate real-world outcomes without that support.
